雪上一枝蒿甲素微透析探针体外回收率研究

[摘要] 该研究建立UPLC-MS/MS测定雪上一枝蒿甲素微透析探针回收率的分析方法,采用浓度差法(增量法,减量法)测定其体外回收率,并考察灌流液的pH,流速,浓度,温度对回收率的影响,以探讨微透析技术用于雪上一枝蒿甲素体内药动学研究的可行性。该试验建立的UPLC-MS/MS分析方法在要求范围内线性良好,色谱的专属性、回收率、精密度等均符合微透析样品的研究要求。增量法和减量法测得回收率无显著差异;相同条件下,探针的体外回收率随着流速的增加而降低,随着温度的升高而明显增加,与探针周围雪上一枝蒿甲素的浓度无关。结果表明,微透析技术可用于雪上一枝蒿甲素药动学研究,反透析法即减量法可用于雪上一枝蒿甲素体内微透析回收率的测定。

[Abstract] To establish UPLC-MS/MS method for determination of the recovery rate of bullatine A microdialysis probe. The concentration difference method(incremental method, decrement method) was used to measure in vitro recoveries, and the effects of perfusate pH value, flow rate, concentration, and temperature on the recovery rate were investigated to explore the feasibility of microdialysis for the pharmacokinetic study of bullatine A. The method of UPLC-MS/MS showed good linear relationship within the required range;the specificity, recovery rate and precision of chromatography met the requirements of microdialysis samples. There was no significant difference in the measured recovery rate between incremental method and decrement method. Under the same conditions, in vitro recovery rate of the probe was decreased with the increase of flow rate, and was significantly increased with the increase of temperature, but was independent of bullatine A concentrations around the probe. The results showed that, microdialysis technology can be used for the pharmacokinetic study of bullatine A, and retrodialysis method (decrement method) can be used for the determination of the in vivo recovery rate of bullatine A microdialysis.

雪上一枝蒿来源于毛茛科植物短柄乌头Aconitum brachypodum Diels,铁棒锤A. Busch及宣威乌头A. subrosullatum H-M.的干燥块根[1],是民间治疗风湿疼痛,关节炎,跌打损伤的要药之一,其主要活性成分为二萜类生物碱类物质,药理实验已证明其具有镇痛、抗炎作用[2]。但因其具有较强的毒性,临床治疗量和中毒量十分接近,用药后常导致中毒,引起心律失常乃至休克[3]。本课题组将雪上一枝蒿研制成微乳经皮给药新制剂,不仅降低了药物的毒副作用,避免肝脏首过效应,同时还提高了药物的生物利用度。目前对于评价经皮给药制剂透皮吸收的传统方法有血药浓度法、离体透皮实验2种方法,这2种方法都无法满足微创,精确定量,局部动态监测等研究要求。

微透析(microdialysis,MD)是近些年临床上发展起来的一项活体生物取样技术,其原理与透析相同,最大的优点是在不干扰生物体的正常生理状态下进行在体(in vivo)、实时(real time)和在线(on line)取样和监测[4]。能在微创的条件下满足定性、定量、定位、微分析、连续取样、动态分析等实验要求,为透皮给药提供了可靠的监测手段[5]。本研究拟使用微透析技术与超高液质谱联用(UPLC-MS/MS)相结合应用于雪上一枝蒿甲素微透析体外回收率的研究,测定其体外回收率并考察其回收率的影响因素,为进一步研究雪上一枝蒿微乳经皮给药制剂的皮肤局部药动学提供可靠的实验依据。

1 材料

微透析系统包括线性探针(CMA30,膜长10 mm,直径0.24 mm),灌注器推进泵(Pump 11 Elite),微量注射针;UPLC-Xevo TQ三重四级杆串联质谱仪(美国Waters);Waters BEH C18色谱柱(2.1 mm×50 mm,1.7 μm);TYXH-Ⅱ型涡旋混合器(无锡沃信仪器有限公司);HC-2062型高速离心机(安徽中科中佳科学仪器有限公司);MTN-2800型氮吹浓缩装置(天津奥特赛恩斯仪器有限公司);AE/240型电子天平(上海梅特勒仪器有限公司)。
