大鼠脑挫伤后碱性成纤维细胞生长因子的表达变化与损伤时间的推断研究

[摘要] 目的 研究大鼠脑挫伤后脑组织中碱性成纤维细胞生长因子(bFGF)时序性表达的规律,探讨bFGF的表达与脑损伤经过时间的关系,旨在为法医实践中推断早期脑损伤时间提供科学依据。 方法 参照Feeney′s法建立SD大鼠脑损伤模型,每组8只将实验动物(n = 64)随机分为8个组(7个实验组,1个对照组),其中实验组依据损伤时间的不同分为1、6、12 h、1、2、3、7 d共7个亚组。应用免疫组化(SABC)法及免疫印迹(Western blot)法检测大鼠脑组织bFGF的含量。 结果 脑挫伤后6 h阳性细胞数逐渐增多,3 d达峰值,7 d已开始下降,但仍高于基础水平。 结论 挫伤后脑组织中bFGF的表达呈现出一定的时序性规律,为早期脑损伤时间的推断提供了实验依据。

The inference study on the change of the expression of basic fibroblast growth factor and the time after brain contusion in rats

[Abstract] Objective To study the temporality expression rule of basic fibroblast growth factor after brain contusion in rats, and to investigate the relationship between the expression of basic fibroblast growth factor (bFGF) and the time after brain contusion, in order to provide scientific evidence for practice of legal medicine to conclude the time of inchoate brain contusion. Methods 64Sprague-Dawley rats were randomly divided into 8 groups (7 of experiment groups and 1 of control group). The rats in experiment group were divided into 7 groups after contusion injury which was performed by the drop method with Feeney's way: 1 h group, 6 h group, 12 h group, 1 d group, 2 d group, 3 d group, 7 d group. Immunohistochemistry and Western blot were used to study the expression levels of bFGF in brain tissue of rats. Results Positive cells increased at 6 hours after brain injury, the most prominent was at third days after contusion injury, and decreased at 7 days after brain injury, but it was more than basal levels. Conclusion There is temporality regular between the expression of bFGF and the time after brain contusion in rats, this regularity can provide experiment evidence for practice of legal medicine to conclude the time of inchoate brain contusion.

碱性成纤维细胞生长因子(basic fibroblast growth factor,bFGF)属于成纤维细胞生长因子家族,是一种存在广泛的多功能肽类生长因子,在细胞的转化、增殖、分化和细胞生长周期中发挥着重要的作用[1]。bFGF广泛存在于中枢及外周神经系统中,对神经组织具有生长分化、维持存活的作用,是有效的神经营养因子之一。本文采用自由落体装置建立大鼠脑挫伤模型,联合采用免疫组化(SABC)法及免疫印迹(Western blot)法两种方法检测该模型中脑挫伤后不同时间段大鼠脑组织中bFGF含量的变化,探讨脑挫伤后bFGF表达的变化与时间的关联性。

1 材料与方法

1.1 实验动物与分组

从山西医科大学实验动物中心购入健康成年SD大鼠64只,体重为250 g左右,雌雄均可,随机分为8组(7个实验组和1个对照组),其中实验组依据损伤时间不同随机命名为1 h组、6 h组、12 h组、1 d组、2 d组、3 d组、7 d组。
